Introducing Neuron Culture Medium

Increase synaptic signaling in iCell® GlutaNeurons with a combination of
BrainPhys™ Neuronal Medium and Wako's Neuron Culture Medium

iCell GlutaNeurons were plated at 30,000 cells/well and maintained in BrainPhys Neuronal Medium until DIV 3. The medium was then replaced and maintained with either complete BrainPhys or complete BrainPhys combined with Wako’s Neuron Culture Medium. Cells were then treated with a dose curve of the potassium channel blocker 4-aminopyridine. Calcium imaging was performed on the FDSS/μCELL (Hamamatsu Photonics K.K.) using Cal-520® AM calcium dye (ATT Bioquest, Inc.). The results suggest that the combined use of these two media facilitates the generation of spontaneous Ca2+ oscillations in iCell GlutaNeurons through the enhancement of maturation of cell function.

Neuron Culture Medium (FUJIFILM Wako Chemicals), when combined with BrainPhys™ Neuronal Medium (STEMCELL Technologies, Inc.), increases synaptic activity as  measured by calcium bursting in iCell® GlutaNeurons.

iCell GlutaNeurons, are a highly pure excitatory neuronal population, expressing AMPA and NMDA that can be modulated using glutamatergic antagonists/agonists, using several types of assays such as calcium signaling and multielectrode arrays. The ability to increase their activity to further develop their maturation in vitro using Wako’s Neuron Culture Medium can enhance the benefits of human neuronal cell modeling as a research tool. Wako’s Neuron Culture Medium is a ready-to-use supplement, capable of inducing rapid neurite outgrowth, using low density cultures and formulated with rat glial supernatant.

iCell GlutaNeurons overcome limitations of existing models by providing the following:

iCell GlutaNeurons were cultured with BrainPhys alone (Panel A) or BrainPhys+Neuron Culture Medium (9:1, Panel B). The images were captured at DIV 3 weeks, following Ca2+ imaging. Cells were stained with Cal-520 AM calcium dye and imaged using a Yokogawa CQ1 confocal cytometer with 10X objective.
