Our User’s Guide instructs to gently transfer the iCell® Cardiomyocytes cryovial contents to a sterile 50 ml centrifuge tube using a 1 ml pipettor and mix by gently inverting the 50 ml tube after addition of iCell Cardiomyocytes Plating Medium. Use of a 50 ml centrifuge tube facilitates suitable mixing and helps to minimize osmotic shock and shear stress and to maximize cardiomyocyte viability. If you choose manual pipetting to resuspend cells or break up clumps, use a wide-bore pipette (e.g. 5 ml) and be careful not to over-pipette the cells.
